ITC (Isothermal titration calorimetry) is a high-resolution method for complete characterization of the basic chemical details of a binding interaction. The method determines a binding constant, reaction stoichiometry, and total reaction energy of two or more compounds (small or large molecules) in solution. Isothermal titration calorimeters do this by measuring the heat that is released or absorbed when molecules interact with each other. Temperature conditions can be controlled to understand the temperature dependence of binding.
The Affinity ITC from TA Instruments | Waters consists of an ultrasensitive calorimeter and a mechanized syringe that reliably delivers a precise quantity of sample into the calorimetric cell containing the target molecule. It is software controlled and experiments are set up, for the syringe to deliver minute volumes of sample at regular intervals. The Affinity ITC can be automated to deliver samples directly from temperature-controlled 96 well plates, for unattended use. Automated cleaning eliminates sample carryover effects, and automated sample loading ensures the sample is delivered to the instrument the same way, every time.
The Affinity ITC allows researchers to characterize the binding of their samples without the use of exogenous tags or dyes, thereby simplifying workflows and reducing errors in biopharmaceutical development. Researchers simultaneously collect stoichiometry data during binding characterization to verify whether a 1-to-1 binding interaction occurs, eliminating the need for other tests. The affinity ITC allows researchers to test their materials under their desired conditions, without reliance on manufacturer-required reagents. Researchers simultaneously collect stoichiometry data during binding characterization and can verify whether a 1-to-1 binding interaction occurs, eliminating the need for other tests.
The affinity ITC provides the specificity of binding, so a fully informed decision can be made on drug candidates from a single experiment. The accompanying NanoAnalyze data analysis software allows users to analyze data in real time, create overlay graphs, and quickly compare drug candidates.
